Technology for Delivery of Poorly Water-Soluble Drugs

Clinical usefulness of many drugs are limited by their poor water-solubility. Akina has developed concepts of hydrotropic polymer (Hytrop®), hydrotropic hydrogel (HyTrogel®), and hydrotropic polymer micelle (HytroCell™).

Hytrops, HyTrogels, and HytroCelles are able to increase the aqueous solubilities of poorly soluble drugs by 2-4 orders of magnitudes. Akina has a series of Hytrops, HyTrogels, and HytroCells that can be used for solubilizing various poorly soluble drugs.

For example, HytroCelles based on PEG-b-poly(2-(4-vinylbenzyloxy)-N,N-diethylnicotinamide) (PEG-b-PVBDENA) can dissolve paclitaxel more than 30 wt% and yet they are stable in aqueous solution for weeks. None of the existing polymeric micelle systems have shown such a high loading efficiency with such a long-term stability. PEG-b-PVBDENA HytroCelles allows parenteral administration of paclitaxel using a very small volume for easy injection. Such HytroyCelles can also be formulated for oral administration of paclitaxel.

PolyVivo: Biodegradable Block CoPolymers

Akina Inc. is currently selling high quality research grade biodegradable copolymers. These polymers are block copolymers of hydrophillic, biocompatible poly(ethylene glycol) (PEG) and biodegradable polymer, such as poly(lactide-co-glycolide) (PLGA), poly(caprolactone) (PCL), and poly(lactide) (PLA). These block copolymers are ideal for delivery of poorly soluble drugs, such as paclitaxel. These polymers can also be utilized for a wide variety of drug delivery platforms including hydrogels (AK08) and other formulations. The polymers shown are of research grade and are excellent tools for developing drug delivery technology.

PolyVitro: Drug Release Media

Also available from Akina is a new product PolyVitro.  This polymer based media modifier generates sink conditions without the characteristic hydrotropic peak which hinders HPLC analysis.  Regular strength PolyVitro has an everolimus solubility of 35 µg/ml while Extra-strength PolyVitro can dissolve everolimus to 50 µg/ml.
